HPLC Column Equivalents: How to Find a Replacement for Agilent, Waters, Thermo, and Supelco Columns

Your validated method calls for a specific column, but it is out of stock, the lead time is six weeks, or the price has jumped. Every chromatography lab hits this problem sooner or later. The good news is that you can often find an HPLC column equivalent if you match the right parameters and verify the results.

This guide explains how to choose a replacement for Agilent, Waters, Thermo Fisher and Supelco (Merck) columns, and when an “equivalent” will not work. If you are still deciding which column type suits your method, start with our complete guide to choosing the right HPLC column.

What Is an HPLC Column Equivalent?

An HPLC column equivalent is a column from a different manufacturer or product line that provides similar retention, selectivity, and peak shape for your method. Two C18 columns can look identical on paper and still separate your sample differently. That is why we say closest match rather than identical. Even the naming can confuse buyers, as explained in our article on C18 vs RP-18 HPLC columns.

Why Labs Look for Column Alternatives

  • Availability: the original column is backordered or discontinued.
  • Cost: a comparable column costs less, especially for routine QC work.
  • Method transfer: a partner lab or a new site uses a different brand.
  • Performance goals: you want faster runs, better peak shape, or a wider pH range.
  • Replacement timing: the old column has reached the end of its life. See signs it’s time for a replacement.

7 Parameters to Match When Choosing a Replacement

1. USP L-Designation

The USP groups columns by packing type. L1 is octadecylsilane (C18), L7 is octylsilane (C8), L11 is phenyl, L10 is nitrile, and L43 is pentafluorophenyl. If your method cites a USP code, start with columns carrying the same code.

2. Bonded Phase (Ligand)

Match C18 to C18, C8 to C8, and phenyl-hexyl to phenyl-hexyl. Switching ligand type changes selectivity significantly. Not sure whether your method needs C18 or C8? Read C18 vs C8 HPLC columns: which one fits your method.

3. Particle Size and Particle Type

Fully porous 5 µm, 3 µm, and sub-2 µm particles behave differently. Core-shell (superficially porous) particles deliver high efficiency at lower backpressure. Keep particle size close to the original, or follow the allowed adjustments in USP <621>. For more detail, see how to choose the right HPLC column particle size.

4. Pore Size

Small molecules usually work well on 80–120 Å. Peptides and proteins need wide-pore packings of 300 Å or more.

5. Carbon Load and End-Capping

Higher carbon load generally increases retention. End-capping affects peak shape for basic compounds. A heavily end-capped column can give very different tailing than a non-end-capped one.

6. pH Stability Range

If your mobile phase runs at pH 9 or 10, you need a hybrid or high-pH-stable column. A standard silica column will degrade quickly.

7. Column Dimensions

Length and internal diameter affect resolution, run time, and solvent use. Keep the length-to-particle-size ratio close to the original method. Our guide to HPLC column dimensions and what they mean explains each number.

Product lines change over time, so always check the manufacturer’s current specifications before ordering. Step-by-Step: How to Select and Verify an Equivalent Column

  1. Collect the original details: brand, product line, part number, USP code, dimensions, particle size, and pore size.
  2. Shortlist two or three candidates with the same ligand and similar specifications.
  3. Use a comparison tool. The USP PQRI column database ranks columns by selectivity similarity, which helps narrow the list.
  4. Run your standard sample on the new column under the original conditions.
  5. Compare the results: retention time, resolution, tailing factor, plate count, and critical pair separation. If retention shifts, check our guide on retention time drift in HPLC. Unexpected extra peaks may be ghost peaks rather than a column problem.
  6. Document the outcome according to your quality system.

When an “Equivalent” Column Will Not Work

  • Critical pair separations: if your method barely resolves two peaks, a small selectivity difference can ruin it.
  • Basic or highly polar compounds: these are very sensitive to silica purity and end-capping.
  • Impurity profiling: a different column can change the elution order of unknown impurities.
  • Strict regulatory methods: some validated methods name a specific column and require formal change control.

Changing a Column in a USP or Pharmaceutical Method

If your method is a pharmacopeial one, check USP <621> allowable adjustments before changing anything. Substituting a different brand usually requires you to demonstrate system suitability (resolution, tailing, precision) on the new column. For validated in-house methods, follow your change-control and revalidation procedures. When in doubt, ask your QA team before swapping the column.

Frequently Asked Questions

Can I replace an Agilent column with a Waters column?

Often yes, if the USP code, ligand, particle size, and pore size match. You must still verify retention, peak shape, and resolution on your own sample.

Is a C18 column from one brand the same as a C18 from another?

No. C18 columns differ in silica purity, bonding density, carbon load, and end-capping, so selectivity can vary.

How do I know if two columns are truly equivalent?

Run the same sample under identical conditions and compare resolution, tailing, and retention. Selectivity databases can help you shortlist candidates first.
